Cross-Talk among RORal and the Rev-erb Family of Orphan Nuclear Receptors
نویسندگان
چکیده
We have cloned Rev-erbfi, a novel isoform of the Rev-erba orphan nuclear receptor. The DNA binding domains of Rev-erba and fl are highly related to each other and to the retinoic acid related orphan receptor (ROR)/RZR subfamily of nuclear receptors. Indeed, we find that all three receptors bind as monomers to the sequence AATGT-AGGTCA. Whereas RORal constitutively activates transcription through this sequence, both isoforms of Reverb are inactive. When coexpressed, both Reverb isoforms suppress the transcriptional activity of RORal. Our data define Rev-erb and ROR/RZR as a family of related receptors with opposing activities on overlapping regulatory networks. (Molecular Endocrinology 8: 1253-1281,1994)

A specific and unusual nuclear localization signal in the DNA binding domain of the Rev-erb orphan receptors.
The orphan receptors Rev-erbalpha and Rev-erbbeta are members of the nuclear receptors superfamily and act as transcriptional repressors. Rev-erbalpha is expressed with a robust circadian rhythm and is involved in liver metabolism through repression of the ApoA1 gene, but no role has been yet defined for Rev-erbbeta. To gain better understanding of their function and mode of action, we characte...

Apolipoprotein CIII (apoCIII) plays an important role in plasma triglyceride and remnant lipoprotein metabolism. Because hypertriglyceridemia is an independent risk factor in coronary artery disease and the presence in plasma of triglyceride-rich remnant lipoproteins is correlated with atherosclerosis, considerable research efforts have been focused on the identification of factors regulating a...
متن کاملRev-erb beta, a new member of the nuclear receptor superfamily, is expressed in the nervous system during chicken development.
We have identified and characterized a new orphan member of the nuclear hormone receptor superfamily in the chicken. This new gene, called Rev-erb beta, exhibits strong homologies with the Rev-erb alpha/ear-1 orphan receptor gene, which partially overlaps the thyroid hormone receptor alpha gene in opposite orientation. We demonstrate that both Rev-erb alpha and Rev-erb beta genes are conserved ...
متن کاملThe orphan Rev-erb nuclear receptors: a link between metabolism, circadian rhythm and inflammation?
Nuclear hormone receptors (NRs) function as ligand dependent DNA binding proteins that translate physiological/nutritional signals into gene regulation. Dysfunctional NR signaling leads to many disorders in reproduction, inflammation, and metabolism. The opportunity to identify novel regulatory pathways in the context of human health and disease drives the challenge to unravel the biological fu...
متن کاملDirect and Indirect Suppression of Interleukin-6 Gene Expression in Murine Macrophages by Nuclear Orphan Receptor REV-ERBα
It is now evident that many nuclear hormone receptors can modulate target gene expression. REV-ERBα, one of the nuclear hormone receptors with the capacity to alter clock function, is critically involved in lipid metabolism, adipogenesis, and the inflammatory response. Recent studies suggest that REV-ERBα plays a key role in the mediation between clockwork and inflammation. The purpose of the c...
